Laboratory of Drug Metabolism and Toxicology

The overall goal of the research program is to enhance understanding of the molecular and cellular mechanisms of bioactivation and detoxification of drugs and endogenous chemicals, and how various patient-specific factors affect drug disposition, efficacy, and toxicity.

A) Regulation of Drug-Metabolizing Enzymes Expression and Function
Drug-metabolizing enzymes are expressed in various tissues, but most of them are present predominantly in the liver. These large diverse groups of enzymes play a significant role in the bioactivation and detoxification of drugs and other chemicals. Studies are ongoing to characterize a select group of human cytosolic drug-metabolizing enzymes (e.g. aldehyde oxidases, sulfotransferases) in health and disease, and identify factors that regulate their expression and function.

B) Pharmacological and Toxicological Consequences of Nuclear Receptor Modulation
Nuclear receptors are DNA-binding transcription factors that regulate the expression of a broad array of genes involved in various physiological and pathophysiological processes, including metabolism and transport of drugs and other chemicals. Current studies include identifying drugs and other chemicals that modulate the function of nuclear receptors and how they affect the detoxification processes of endogenous chemicals (e.g. bile acids).
